Bullseye Biosciences is at the vanguard of directed evolution, innovating on therapeutic discovery for critical targets in oncology, immunology, and metabolism. The “Archer” platform has been established from the foundational work of our academic co-founder, Dr Chory at Duke. We seek to deploy this technology against intracellular protein-protein interactions (PPIs) which are too large for small molecules and inaccessible to biologics. PPIs are extremely well validated drivers of human disease whose potential as drug targets remains a high unmet need

Your expertise in directed evolution will not just advance our research, but also contribute significantly to the development of life-changing treatments. Role Description:

Come join us as a Directed Evolution Scientist in building a new generation of drugs against historically challenging targets using our directed evolution discovery engine! This role will be instrumental in building our synthetic biology capabilities and executing our screening campaigns against challenging targets. The Archer screening platform and subsequent hit testing relies on careful design of reporter “gene circuits” with multiple inducible components for precise control of the system. The Directed Evolution Scientist would help design, validate, and calibrate these systems for high-throughput evolution screening. Working with the head of research and automation scientists, this scientist would be accountable for the design and validation of novel gene circuits against challenging human disease targets

The directed evolution scientist would work with our automation scientists and our academic founder’s lab to implement these circuits on high-throughput continuous evolution systems. They would also have the opportunity to continue supporting these targets through early drug discovery efforts. As a member of a small start-up everyone makes major contributions to the direction and success of the company!

Key Qualifications and responsibilities:

  • Experience in PACE or similar directed evolution and 2-hybrid techniques in E Coli systems
  • Molecular biology laboratory techniques including bacterial culture, PCR, cloning, protein blot analysis, assay development (western blot, ELISA, luminescence), and phage propagation
  • Expertise with plasmid/construct design and cloning techniques. Construct design and tracking using Geneious and Benchling. Sequencing by Sanger for confirmation.
  • Understanding of plasmid components, such as inducible promoters and RBS optimization
  • Attention to detail and accuracy in laboratory work including familiarity with ELN programs
  • Strong analytical and problem-solving skills
  • Proficiency in data analysis and graphing software, including Graphpad, Spotfire, and Excel
  • Experience with RNA/DNA NGS and automation is a plus
  • Ability to work independently and as a team
  • Good organizational and time management skills
  • PhD with at least 2 years of experience, or a Master's degree with at least 4 years of experience, in synthetic biology or a related field
